Yes, once registered, you can immediately configure DNS resolution creating a profile in the account for the domains. The resolution process may take up to 24hrs to effect, however in most of the case, it is only require few minutes to effect, unless the TLD requires some verification process to identity the identity of the domain holder. 

If you don’t renew your domain on time, there is normally a grace renewal period of up to 30-day, then domains will line up for de-registration. Normally domain owner will still have a last chance to redeem an expired domain name, however at this state one may need to pay extra "Redemption Fee". If no action is taken after all the grace period, the domain might be secured by someone else and it will effect your website resolution and workability.
